ABSTRACT

Objective:To evaluate the in vitro cross-neutralization of serum antibodies in human and mice immunized with inactivated SARS-CoV-2 vaccine against Delta and Beta variants. Methods:Human serum samples after a second and a third dose of inactivated SARS-CoV-2 vaccine and mouse serum samples after a two-dose vaccination were collected. The neutralizing antibodies in the samples against SARS-CoV-2 strains of prototype, Delta and Beta variants were detected using micro-neutralization assay in biosafety level Ⅲ laboratory. The seroconversion rates and geometric mean titers (GMTs) of antibodies were calculated.Results:The seroconversion rates of antibodies in human serum samples against different SARS-CoV-2 strains were all above 95%. After two-dose vaccination, the GMTs of neutralizing antibodies against the prototype, Delta and Beta strains were 109, 41 and 15, respectively. The GMTs decreased by 2.7 folds and 7.3 folds for the Delta and Beta variants as compared with the prototype strain. After the booster vaccination, the GMTs of neutralizing antibodies against the prototype, Delta and Beta strains were 446, 190 and 86, respectively. The GMTs of neutralizing antibodies against Delta and Beta variants decreased by 2.3 folds and 5.2 folds as compared with that against the prototype strain. The seroconversion rates of antibodies against different SARS-CoV-2 strains in mouse serum samples were all 100%. The GMTs of neutralizing antibodies against the prototype, Delta and Beta strains were 2 037, 862 and 408, respectively. The GMTs decreased by 2.4 folds and 5.0 folds for the Delta and Beta variants.Conclusions:Inactivated SARS-CoV-2 vaccine could induce a certain level of neutralizing antibodies against Delta and Beta variants in both human and mouse models. Moreover, a third dose of vaccine induced higher levels of neutralizing antibodies against Delta and Beta variants in human. This study provided valuable data for the clinical application and protective evaluation of the inactivated SARS-CoV-2 vaccine.

ABSTRACT

Toll-like receptor 3 (TLR3) is a member of the TLR family, mediating the transcriptional induction of type I interferons (IFNs), proinflammatory cytokines, and chemokines, thereby collectively establishing an antiviral host response. Studies have shown that unlike other TLR family members, TLR3 is the only RNA sensor that is utterly dependent on the Toll-interleukin-1 receptor (TIR)‍-domain-containing adaptor-inducing IFN-‍β (TRIF). However, the details of how the TLR3-TRIF signaling pathway works in an antiviral response and how it is regulated are unclear. In this review, we focus on recent advances in understanding the antiviral mechanism of the TRIF pathway and describe the essential characteristics of TLR3 and its antiviral effects. Advancing our understanding of TLR3 may contribute to disease diagnosis and could foster the development of novel treatments for viral diseases.

ABSTRACT

Bluetongue virus (BTV) causes Bluetongue (BT) of ruminants vectored by culicoides midges. It is also a classic model for studying the release mechanism of non-enveloped virus. This review begins with the infection and assembly of BTV, then summarizes the advances of different ways of releasing BTV. This includes BTV-induced autophagy and the release as extracellular vesicles via multivesicular bodies, BTV-induced apoptosis and the lytic release, as well as different pathways of release through budding via plasma membrane. The regulatory mechanisms of NS3 which is a key non-structural protein during the release of BTV are also discussed, providing a basis for further understanding the molecular mechanisms underpinning the infection, proliferation and release of BTV.

ABSTRACT

Objective:To investigate the relationship between snoring and the risk of cardiovascular and cerebrovascular events.Methods:By searching PubMed, Embase, the Cochrane Library, China National Knowledge Infrastructur, Wanfang Database, VIP Chinese journal database and Chinese biomedicine databases from the establishment to June 10, 2019, relevant domestic and foreign literature, extract data and apply Newcastle-Ottawa Quality Assessment Scale (NOS)method to quality evaluation, and finally integrate the data and analyze with Stata12.0 software.Results:A total of 11 articles and 145 267 participants met the inclusion criteria.Meta-analysis results showed that the correlation strength and 95% CI of snoring with cardiovascular and cerebrovascular events and stroke risk were 1.10 (1.03-1.17) and 1.26 (1.11-1.43)respectively , and all of them had statistical significance.Conclusion:Snoring is an independent risk factor for the risk of cardiovascular events and is more closely linked to stroke.

ABSTRACT

Objective To investigate the expression changes of soluble urokinase-type plasminogen activator receptor(suPAR)and soluble triggering receptors expressed by myeloid cell-1(sTREM-1)in serum of children with primary nephrotic syndrome(PNS)and their clinical significance.Methods A total of 92 cases of newly diag-nosed PNS children were selected in Central Hospital of Yidu Affiliated to Weifang Medical College from June 2014 to September 2016.According to presence or absence of acute tubular necrosis,they were divided into acute renal injury group(27 cases)and non-acute renal injury group(65 cases).According to pathology type,they were divided into mesangial proliferative glomerulonephritis(30 cases),focal segmental glomerulosclerosis(23 cases),membranous ne-phropathy(18 cases),minimal change disease(14 cases)and membrane proliferative glomerulonephritis(7 cases).In the same period,45 healthy children were selected as the healthy control group.The clinical data were collected.The serum levels of suPAR and sTREM-1 were measured by adopting enzyme-linked immunosorbent assay(ELISA). Results The levels of total cholesterol(TC),triglycerides(TG),uric acid(UA),urinary protein/creatinine,24 h urinary protein,urinary N-acetyl-β-glucosaminidase(NAG)and β2-microglobulin(MG)in children with PNS were higher than those in the healthy control group,while serum albumin(ALB)was lower than that in the healthy con-trol group,and the differences were statistically significant(all P<0.05).The serum levels of suPAR and sTREM-1 in PNS patients were(133.09 ± 62.48)ng/L and(79.29 ± 34.68),respectively,which were significantly higher than those in the healthy control group[(31.11 ± 11.61)ng/L and(25.08 ± 8.10)ng/L](t=51.714,49.435;all P=0.000).The serum levels of suPAR and sTREM-1 in acute renal injury group were(188.82 ± 32.21)ng/L and (109.11 ± 24.78)ng/L,respectively,which were significantly higher than those in non -acute renal injury group [(75.96 ± 28.69)ng/L and(52.23 ± 14.07)ng/L]and healthy control group[(31.11 ± 11.61)ng/L and (25.08 ± 8.10)ng/L](F=16 739.607,10 487.256,all P=0.000).The serum levels of suPAR and sTREM-1 in children with focal segmental glomerulosclerosis and membrane proliferative glomerulonephritis were higher than those with minimal change disease,membranous nephropathy and mesangial proliferative glomerulonephritis,and the differences were statistically significant(all P<0.05).Pearson correlation analysis results showed that the serum levels of suPAR and sTREM -1 were positively correlated with TC,TG,urinary protein/creatinine,24 h urinary protein, urinary NAG and β2-MG(all P <0.05),while negatively correlated with ALB(P <0.05). Conclusions The serum levels of suPAR and sTREM-1 are elevated in children with PNS,and which are related with acute renal injury and pathological type,which can reflect the degree of renal tubular disease and kidney function to a certain extent.

ABSTRACT

AIM To explore the effects of Xuebijing Injection (Carthami Flos,Salviae miltiorrhizae Radix et Rhizoma,Paeoniae Radix Rubra,etc.) on serum CRP,TNF-α and IL-6 levels in patients with severe acute organophosphorus pesticide poisoning (AOPP).METHODS Seventy-two cases of patients with AOPP treated from Jan 2014 to Mar 2016 in the emergency department of our hospital were randomly divided into two groups,the control group treated with atropine,pralidoxime chloride and conventional treatment,and the observation group combined with Xuebijing Injection.The dynamic changes of serum CRP,TNF-α and IL-6 levels,and clinical curative effects were compared between the two groups.RESULTS The serum CRP,TNF-α and IL-6 levels in the two groups were decreased in turn before the treatment,at the 3rd,7th days after the treatment,and the serum CRP,TNF-α and IL-6 levels in the observation group were significantly lower than those in the control group;the dosage of atropine,time of cholinesterase activity recovered 60% and hospital stay in the observation group were significantly less than those in the control group;the rebound rate of the observation group was lower than that of the control group.The acute respiratory distress syndrome (ARDS) rate in the observation group was significantly lower than that in the control group.All the differences had statistical significances (P < 0.05).CONCLUSION Xuebijing Injection can effectively inhibit the inflammatory response,reduce the incidence of complications,shorten the course,and improve the clinical efficacy in the treatment of patients with AOPP.

Objective@#To investigate the correlation between the expression of PD-1, PD-L1 and clinicopathologic parameters in triple negative breast carcinoma (TNBC).@*Methods@#Samples from 151 patients with TNBC and 65 cases of other breast carcinomas (non-TNBC) were examined for PD-L1 and PD-1 expression by immunohistochemical staining on tissue microarray.@*Results@#The expression of PD-L1 and PD-1 in the tumor cells and interstitial lymphocytes in TNBC was significantly (P<0.05)higher than that in non-TNBC.In TNBC, the expression rates of PD-L1 in the cancer nests and stroma were 16.6%(25/151) and 25.2%(38/151)respectively.The former was positively correlated with tumor histological grade and lymph node metastasis (P<0.05), and the latter only with tumor histological grade (P<0.05). The expression rate of PD-1 was 27.2% (41/151), and correlated with PD-L1 expression in stromal tumor-infiltrating lymphocytes (P<0.05).@*Conclusion@#The expression of PD-L1 in TNBC is significantly correlated with high-grade histology, lymph node metastasis and PD-L1 expression in stromal tumor-infiltrating lymphocytes.

ABSTRACT

OBJECTIVE:To establish a method for the content determination of calceolarioside A in the Leaves of Fraxi-nus mandschurica. METHODS:HPCE was performed on the capillary of uncoated fused silica capillary,buffer solution of borate buffered saline(20 mmol/L,pH 9.3),separation voltage of 20 KV by pressure injection(injection pressure was 50 bar with 3 s), detection wavelength was 350 nm,and column temperature was 30 ℃. RESULTS:The linear range of calceolarioside A was 50-250μg/ml(r=0.999 7);RSDs of precision,stability and reproducibility tests were less than 3%,recovery was 97.48%-102.08%(RSD=1.12,n=6). CONCLUSIONS:The method is simple,good reproducibility,and can be used for the content determination of calceolarioside A in the Leaves of F. mandschurica.

ABSTRACT

PURPOSE: The aim of this study is to explore signal transducer and activator of transcription 3 (STAT3) expression in breast cancer and to analyze the detailed mechanism that STAT3 contributes to the progression of breast cancer. METHODS: We retrospectively analyzed the clinicopathologic characteristics and overall survival (OS) of 140 breast cancer patients after curative surgery, and detected STAT3 expression, phosphorylated STAT3 (pSTAT3) expression, Ki-67 expression, vascular endothelial growth factor (VEGF)-C and -D expression in breast cancer tissues, and adjacent nontumor tissues. Survival analysis and relationship analysis were adopted for demonstrated the important mechanism of STAT3 contribution to progression of breast cancer. RESULTS: STAT3 expression, pSTAT3 expression, Ki-67 expression, VEGF-C expression, and VEGF-D expression in breast cancer tissues were significantly higher than those in adjacent nontumor tissues, respectively. With survival analysis, only number of lymph node metastasis (N stage) was identified as the independent predictors of the OS of breast cancer patients. Besides, we demonstrated there was the most prominent correlation between STAT3 expression and lymph node metastasis in breast cancer tissues by using the multinominal regression method. CONCLUSION: STAT3, a poor survival biomarker potential association with lymph node metastasis, was suitable for predication the OS of breast cancer patients after curative resection.

BACKGROUND:Isokinetic test system has been widely used to evaluate the muscle function levels Whereas.the reports concerning effect of sport training on the development of muscle strength of hip joint in college students using isokinetic testing are few.OBJECTIVE:To assess the extensor and flexor muscle strength of hip joints in college students with isokinetic testing system METHODS:Totally 40 volunteers,including 20 male and 20 female students were selected.Kinitech isokinetic testing system,produced by Australian Kylingk Company,was used to test the flexion and extension muscle of hip ioints as the sequence of centripetal followed by centrifugal.The test speed was 60(°)/s as slow speed,180(°)/a as middle speed,240(°)/s as high speed.Peak torque(PT)and relative values of extension and flexion muscles,total power(TP)and relative total power were measured.RESULTS AND CONCLUSION:At the respective speed,PT and relative values of female students were greater than that of female students(P<0.01).Atthe speed of 60,120 and 240(°)/s,flexor PT was smaller than that of extensor PT in male students,which was opposite in the female students,but the difference had no significance(P>0 05).At the respective speed.TP of males were greater than that of female students(P<0.01).In male students,TP and relative values of flexor were smaller than extensorat speed of 60(°)/s and 120(°)/s(P<0.01),and had difference at speed of 240(°)/s(P<0.05).Infemale students,TP and relative values had significant difference between flexor and extensor at speed of 60(°)/s(P<0 01)and at speed of 120(°)/sand 240(°)/s(P<0 05).At the respective speed,PT and relative values,TP and relative values in male students are all greater than females.These values decrease with the speed increasing.At the respective values,values of male students are bigger than females.

BACKGROUND:As an effective method to evaluate the muscle functional level in humans,isokinetic testing system is applied in more and more researches.However,reports remain few in which isokinetic muscle strength test is used to study the effect of movement training on the development of muscle groups in teenagers.OBJECTIVE:To make a comparison and analysis on the muscle strength of knee joints between the two different teenager populations of the same age,namely,the one with jump testing and the one without.DESIGN,TIME AND SETTING:A contrast observation was performed at the Laboratory of Hebel Physical Education Science Institute between October and Decamber in 2008.PARTICIPANTS:The 20 volunteers without sport training were selected randomly from Hebei Normal University Affiliated Middle School in Shijiazhuang,and were classified as the control group;the other 20 volunteers with high jump and long jump training were randomly selected from Shijiazhuang Sports School,and were used as the training group.All participants are males.METHODS:Cybex-6000 isokinetic testing system was used to test the knee joints of all participants,with the concentdc contraction going first and then the eccentric contraction.The slow,middle and high testing speeds were 60(°)/s,120(°)/s and 240(°)/s respectively.MAIN OUTCOME MEASURES:Relative peak torque(PT)values of flexor and extensor groups in knee joints.RESULTS:All 40 volunteers were involved in the result analysis.Compared with the control group,knee joints in the training group showed much higher flexor and extensor PT values during the concantric contraction and the eccentric contraction at the speed of either 60(°)/s or 120(°)/s or 240(°)/s,and the difference between the two groups was of significance(P<0.01).The PT value ratios between flexor and extensor of both the left and the right knee joints were lower in the training group than in the control group at any of the three speeds.CONCLUSION:Jump exercise has a distinctively positive effect on increasing the muscle strength of knee joints in teenagers,while it is limited in the coordinated development of extensor and flexor,leading to the decreased ratios between the two which may be caused by the inappropriate training method.

BACKGROUND: Nowadays, more and more people regard the isokinetic training as a method for strength training and rehabilitation of muscle injuries of athletes. But it is rare in study of rehabilitation following joint injury via isokinetic training. OBJECTIVE: To explore the advantage of isokinetic training in rehabilitation, further more, to provide the scientific basis to rehabilitation of muscle injuries. DESIGN, TIME AND SETTING: The contrast observation was performed at the Laboratory of Institute of Physical Education, Hebei Normal University between March and May 2008. PARTICIPANTS: Two female college basketball athletes whose knee joints were injured. METHODS: The Kinitech isokinetic testing system (Kylingk, Australia) was used to test the flexion and extension muscles of knee joint of 2 athletes and contrasted the values of flexion and extension muscle after rehabilitation training, damage side and healthy side. All data were analyzed to observe the effects of strength training. MAIN OUTCOME MEASURES: Peak torque (PT) values of flexor and extensor in knee joints. RESULTS: After 8 weeks isokinetic training, the strength index of two subjects had increased in certain degree both at the 60 (?)/s and 240 (?)/s. The PT values showed that the increasing range of flexor was greater than that of extensor in the 1st subject, but the circumstance was opposite in the 2nd subject. The index of BP, STW, and AP was greatly increased, as well as work fatigue. CONCLUSION: It is helpful to increase the muscle strength and promote the rehabilitation via isokinetic training system. The isokinetic strength training has the positive effects on rehabilitation of muscle injuries.

ABSTRACT

AIM: To study the development laws between isokinetic muscle strength and age increase via test flexion and extension muscle of knee joints in 216 ordinary juvenile boys aged from 9 to 20. METHODS: Kinitech isokinetic strength test was used to study the flexion and extension muscle of both sides’ knee joints at the speed of 60 (?)/s. RESULTS: The peak torque of muscles increased with the increasing ages, especially in extension muscles, which is bigger than flexion muscles (P 0.05). The ratio of flexion and extension muscles (F/E) decreased with age increasing, which a larger fluctuation during aged 9-13, especially obvious had decreased in 13-20 ages. CONCLUSION: The muscle strength and power all increases with the increasing ages, while the F/E value decrease in 9-20 boys, which demonstrates the flexion and extension muscle developed out of balance.

BACKGROUND:In isokinetic testing researches,the effect of sport training on the development of teenager muscle groups is less reported. OBJECTIVE:To assess the muscle strength of shoulder,hip and knee joints in different teenager populations with isokinetic testing system. DESIGN,TIME AND SETTING:The contrast observation was performed at the laboratory of Institute of Physical Education,Hebei Normal University between October and December 2006. PARTICIPANTS:Control group included 30 volunteers without sport training randomly selected from four middle schools in Shijiazhuang,15 male and 15 female;training group included 30 volunteers with sprint training randomly selected from Shijiazhuang Sports School,15 male and 15 female. METHODS:We used Kinitech isokinetic testing system(Kylingk,Austrilia) to test the shoulder,hip and knee joint of the 60 participants according to 3 different angular velocities:60( ?) /s,120(?) /s and 240( ?) /s. MAIN OUTCOME MEASURES:Peak torque(PT) values of extensor and flexor groups in shoulder,hip and knee joints. RESULTS:The 60 volunteers were all included in the result analysis. ABSTRACT

OBJECTIVE To investigate and control the risk factors about nosocomial infection in cerebral hemorrhage patients.METHODS Totally 986 hospitalized patients with cerebral hemorrhage in our hospital from Jan 2002 to Dec 2005 were studied retrospectively.RESULTS It showed that the incidence of nosocomial infection rate was 32.05%,case infection rate was 44.62%.The nosocomial respiratory tract infection was the highest(66.36%),the second one was the urinary tract(20.68%).We also found that the risk factors prone to nosocomial infection were aging,long hospitalization,respiratory machine application,respiratory tract inicison,indwelling catheter and coma.CONCLUSIONS Nosocomial infection is still a high frequent complication in cerebral hemorrhage.It is suggested that there be urgent need for treating the underlying disease and reducing the risk factors,then can reduce the development of infection in patients with cerebral hemorrhage.

ABSTRACT

cholesterol 7α-hydroxylase gene ( CYP7A 1 ) plays a key role in the catabolism of cholesterol into bile acids. To investigate whether the A-204C polymorphism in CYP7A1 gene affects the gene expression,using luciferase as the reporter gene, four recombinants were constructed by inserting forward or reverse sequence with A or C allele at the polymorphism site into the promoter-less vector pGL3-basic. The constructs were then transfected into four cell lines and the luciferase activity of each expression vector was examined by dual luciferase reporter gene assay system. The results showed that activities of the forward sequence of both genotypes were higher than that of reverse sequence. Promoter activity of the recombinants with A allele was about one third lower than that with C allele. According to the analysis with TRANSFAC database, there may exist a Zic3 binding site when there is the C allele at -204. Our study indicates that the A-204 C polymorphism in CYP7A1 promoter region decreases its promoter activity and thus represses the gene expression, possibly due to the lack of a potential Zic3 binding site.
